I think Jim Aparo gets my vote by a narrow margin some others already mentioned.

Brian Bolland's KILLING JOKE has a lot of things I don't care for in my
comics these days, but a badly drawn Joker's not one of them. It's
always been my favorite. However, a "little known" artist who came to
the states from England, via Canada pulled a little gem out of his
pencil and shared it in his gallery...

That Joker is the creepiest vie ever seen. I'd be legitimately scared of
that man were he real.

The original, heavier Joker, Dick Sprang.

The later, skinny Joker, Neal Adams.

And an honorable mention to Bruce Timm for the animated version he first designed.

Neal Adams.  "The Joker's Five Way Revenge" is my single-favorite issue of Batman- cool to see the opening page image posted in this thread.

So many good choices, including all of those mentioned already.

I personally will go with Adams' Joker; but that's a very narrow lead over the rest.
