Calvin and Hobbes by a landslide.  Something to be said for quitting while you're still at the top of your game.

Prior to that it was Dennis the Menace.  Definitely some similarities there.

The only still running newspaper strip I read with any regularity is Dick Tracy. And I read it online.

When I was a young teen, Funky Winkerbean. 
An older teen, Doonesbury then the Far Side. 
As a 20-something, Calvin & Hobbs. Not sure anything could surpass Calvin & Hobbs.

My comic strip reading is in two parts, one on each side of the Atlantic.

As a child in England my favorite was Rupert Bear. Once we crossed to Canada (and the accompanying exposure to American strips) I was won over by Pogo.

THE FAR SIDE — hands down. Goofy yet sophisticated; low brow intermingled with high brow. Also: sentient cows. I get a laugh out of it maybe 95/100 times. 

As far as favorite strips with multiple panels, I’d have to say … ZIGGY.
